SMC is administered using co-blistered sulfadoxine-pyrimethamine (SP) together with amodiaquine (AQ). This combination was chosen due to its ability to provide relatively long-lasting protection.

SPAQ is currently dosed at monthly intervals for three to five months during the rainy season. This combination is a successful example of the re-purposing of two antimalarial drugs.
